Selenium and Vitamin E modulatory effects on
genomic DNA Double-Strand Breaks induced by
chemical carcinogens in acute treatment in Wistar

Worldwide literature data show protective
effects of organic Selenium and Vitamin E against
liver chemical injuries. Previous results
obtained in our laboratory by radioisotope
(3H-Thymidine) uptake method indicate that
organic Selenium salts in combination with
Vitamin E has a protective effect on hepatocytes
treated with 2-Acetylaminofluorene (2-AAF). In
order to further explore the modulatory effects
of antioxidants on hepatocytes treated with
DSB-inducing agents, we have investigated the
patterns of DNA fragmentation induced by 2-AAF in
acute treatment on Wistar rats by means of CHEF
III and densitometric analysis of the resulting
gels. We hereby report the results of this work
and that the fragmentation patterns obtained
correlate with previous radioisotopic uptake
results, indicating that organic Se salts and
Vitamin E have indeed a protective effect against
2-AAF-induced DSB in rat hepatocytes.
