Abstract
The study presents an integrated micro
Reverse Transcription Polymerase Chain Reaction
(RT-PCR) system for molecular diagnosis of
microorganisms. The developed system has
integrated biosample pretreatment devices and
RT-PCR devices and by using superparamagnetic
beads can detect viruses with a higher
sensitivity, when compared with large-scale
machines available in the market. The target
viruses were firstly captured by specific
antibody-conjugated magnetic beads, and trapped
in a magnetic field generated by microcoils.
Then, RNA of the virus were extracted and
translated to cDNA, followed by amplification
process using the micro temperature control
module. The integrated micro system can perform
automatic mixing, transportation and
amplification of virus samples and could lead to
a crucial platform for future biological and
medical applications.
Experimental Process
Typical thermal cycles generated by the micro
RT-PCR chip. The temperature variation was less
than 0.2?.The heating rate is 20?/sec, cooling
rate is 10?/sec.
