Title: DETECTION OF DENATURATION OF PROTEIN AT LOW TEMPERATURE USING FTIR

1
DETECTION OF DENATURATION OF PROTEIN AT LOW
TEMPERATURE USING FTIR
  • BY VIKAS W. KAPOOR
  • CHEMICAL ENGINEERING DEPARTMENT
  • SYRACUSE UNIVERSITY
  • SYRACUSE

2
CONTENT
  • Denaturation of protein
  • Denaturation occurring at lower temperature
  • Its detection using FTIR
  • Results and conclusion

3
Denaturation of proteins
  • What is denaturation of protein?
  • It is the change taking place in protein
structure in such a way that they are no more
functional. These changes are mainly dependent on
    their secondary and tertiary structures.

4
Structue of protein
a. Primary Structure b. Secondary Structure c.
Tertiary Structure d. Quaternary Structure
5
Effect of denaturation on properties of protein.
  • Decrease in solubility
  • Altered water binding capacity
  • Loss of biological activity
  • Improved digestibility
  • Increased intrinsic viscosity
  • Inability to crystallize

6
Techniques to detect protein denaturation.
  • Fourier Transform Infrared Spectroscopy(FTIR)
  • Differential Scanning Calorimetry(DSC)
  • Circular Dichroism(CD)
  • Birefringence
  • Optical opacity

7
Denaturation at lower temprature
  • Need of this study
  • Reasons of denaturation at lower temperature on
    proteins
  • Study of AT - 1 Dunning prostrate tumor cells
    done at control condition(normal condition),
-20C and - 80C by using FTIR to see the
denaturation of protein occurring at freezing
    temperatures.
  • It was observed using FTIR that protein
denaturation was observed significant at - 80C
    than -20C
