2The Question
- Compare the action of Valium and Anectine on the
somatic motor pathway, focusing on their effects
on skeletal muscle and/or neuron membranes.
3Valium (Diazepam) is often prescribed to relieve
muscle spasm without interfering with muscle
function. Anectine (Succinylcholine) is a rapid
skeletal muscle relaxant, and is used as an
adjunct to general anesthesia (during surgery).
We will now compare the action of Valium and
Anectine on the somatic motor pathway, focusing
on their effects on skeletal muscle and/or neuron
membranes.
4Valium (Diazepam)
- Valium enhances and facilitates the
neurotransmitter GABA in the Central Nervous
System - GABA an inhibitory neurotransmitter that
mediates pre- and post-synaptic inhibition in all
of the CNS, esp. hypothalamus limbic system and
upper motor neurons. - So, what does Valium do?
- Valium modifies the GABA A receptors so the
receptor is more sensitive/responsive to the
neurotransmitter GABA. - Therefore, more GABA inhibiting the neuron makes
it harder to generate an action potential.
5GABA Receptor
- Normal action of GABA at receptor is to allow
influx of Cl-, hyperpolarizing the cell - Several ligands may affect cell function by
binding to different subunits of the GABA receptor
6Anectine (Succinylcholine Chloride)
- Anectine combines with cholinergic (nicotinic)
receptors at the motor end plate and causes
muscle cell depolarization. - Anectine acts like acetylcholine. It attaches to
the receptors at the motor end plate. - Once it attaches it opens Na channels (like
ACH), and depolarizes the cell. AT FIRST - Once it has attached to the receptor it lasts
around 4 to 10 minutes before it is metabolized
by plasma pseudocholinesterase - It prevents ACH from having an effect on the
receptor and therefore cant stimulate the muscle
cell because the membrane is refractory - Initially fasciculations occur upon
administration with rapid termination.

8The Overall Effects
- Valium enhances GABAs ability to inhibit the
somatic motor pathway. Helping GABA to
GETRDONE - Meaning less action potentials are conducted
resulting in decreased muscle contractions. - Anectine blocks Ach at the receptor and
completely inhibits Ach from getting to the
muscle cell. - Meaning NO muscle movement
