2
Objectives
  • To develop an undergraduate student laboratory
that emphasizes current nanoscience research in
    biomedical engineering
  • To introduce the theory and use of atomic force
microscopy (AFM) with the Nanosurf? EasyScan 2 AFM

3
Potential experiments
  • Attempted samples
  • Diatomaceous earth
  • Nickel nanowires
  • LBL assembly of M13 phage
  • EPROM
  • Nanopatterned PCL
  • Gold nanoparticles
  • Criteria
  • Use current nanoscale techniques
  • Able to be synthesized by the students
  • Can be analyzed with the AFM

4
What didnt work
  • Diatomaceous earth
  • Diluted in 1x TAE Mg2 buffer and air-dried on
    mica for 3 days
  • Nickel nanowires
  • Diluted in 75 EtOH and air-dried on mica for 3
    days
  • Layer-by-layer assembly
  • LPEI would not dissolve! Nature paper seems bogus

5
What did work
  • EPROM
  • Broke IC open to expose chip
  • Nanopatterned PCL
  • Soft lithography using 350 nm patterned gratings
    embossed on polycaprolactone (PCL)
  • Gold nanoparticles
  • Synthesized using HAuCl4 and citric acid and
    ascorbic acid

6
EPROM
7
Nanopatterned PCL
8
Gold Nano(micro?)particles - Ascorbic Acid
9
Gold Nano(micro?)particles - Ascorbic Acid
10
Gold Nano(micro?)particles - Ascorbic Acid
11
Gold Nano(micro?)particles - Citric Acid
12
Lab Manual
  • Discussion of AFM Theory
  • Introduction to soft lithography and usage of
    nanopatterned topography in tissue engineering
  • Example of gold nanoparticles as a therapeutic
    agent - photothermal ablation
  • Protocol for AFM operation to view EPROM, PCL
    nanopatterns, and gold nanoparticles
  • Protocol to make PCL nanopatterns
