WHAT IS IGEM?
  • International Genetically Engineered Machine
  • Having received a kit of biological parts from
the iGEM Registry, we start to build our
    biological systems over the summer.
  • The perspective of the Jamboree in November
at MIT and the project design is exceptionally
motivating, in addition to the fact that this is
    a good learning experience.

THE PROJECT WET LAB PART
5
THE PROJECT WET LAB PART
  • Cloning Parts
  • ? using the BioBrick protocol of Ginkgo
    Bioworks

6
THE PROJECT MAKING A PART
7
THE PROJECT WET LAB PART
  • The BphP / PpsR system

8
THE PROJECT MODELING PART-AIM
Goal Enhance the binary effect of the light
switch
  • Molecular dynamics (MD) simulation of the
photosensitive protein domain on computer for two
    protein states
  • The light activated state
  • The ground state (protein without light
    excitation)

Study of both state stability (statistical
analysis)
Determination of amino acids that could be
mutated to enhance stability of light activated
state
Mutation of the amino acids and test the switch
in vivo
9
THE PROJECT MODELING PART-PRELIMINARY RESULTS
  • First nanosecond MD simulation

10
THE PROJECT APPLICATIONS
  • Merck-Serono interests
  • using light to induce gene transcription in
    mammalian cells
