Major stages of Semi-Conservative DNA Replication
1. Unwind separate strands
Helicase, Gyrase, SSBPs
2. Initiate building of new strands by priming
Primase RNA primers
3. Continue building new strands using
complimentary base pairing rules
4. Double check new strands to make sure
everything is honky-dory
DNA Polymerase I, Ligase
5
The lagging strand is synthesized
discontinuously. Primase synthesizes a short RNA
primer, which is extended by DNA polymerase to
form an Okazaki fragment.
After the RNA primer is replaced by DNA (by
another DNA polymerase, not shown) DNA ligase
joins the Okazaki fragment to the growing strand.
Single-strand binding proteins stabilize the
unwound parental DNA.
Helicases unwind the parental double helix.
