Title:

The General Toxicological Study of Traditional Chinese Medicine Drug -- Mandala

Description:

We choose mice as the experimental animals to study the acute toxicity and chronic toxicity of Datura stramonium. The results show that, the mice can tolerate greater than 4 g / kg and less than 16 g / kg of Datura stramonium. The median lethal dose: LD50=6.82 g / kg, 95% confidence interval for [5.71, 8.59]. Large and long-term administration will damage liver and kidney. Mandala is not suitable for long-term administration in clinical administration. if it is really necessary to give long-term administration, we should strictly control the dosage, and monitor the liver function and renal function – PowerPoint PPT presentation

  • In 1970, Chinese herbal anesthesia, like Datura
flower was rediscovered. Its main effects are to
relieve cough, asthma, pain and spasmolysis. For
the use of asthma cough, abdominal cold pain,
    rheumatism, chronic infantile convulsion,
surgery. Datura flower belongs to anticholinergic
drugs based on M cholinergic receptor. In recent
years, it makes significant progress in the
clinical application. But toxic Mandala will
cause toxic reactions with improper dosage.

  • Results and analysis
  • 1. Acute poison experiment
  • To determine the toxicity of Datura in mice by
    intragastric administration. The results show
that, the mice can tolerate greater than 4 g / kg
and less than 16 g / kg of Datura stramonium. The
    median lethal dose LD506.82 g / kg, 95
    confidence interval for 5.71, 8.59.

  • 2. Long term poison experiment
  • 2.1. General status and weight growth of animals
  • Animals are reared for 92 days, and mice are
administered 1 time a day. Control group, low
dose group and middle dose group animals keep
their hair smooth, behavior rapid and reaction
quick, but high dose group animals have changed
significantly (P lt 0.05), and significantly
reduce food intake, less dynamic. The results
show that the mice have obvious toxic reaction
    after long-term administration of high-dose
    group.

5
  • 2.2 Animal mortality
  • No animal dies during the experiment.
  • 2.3 Animal liver function
  • Taking the mouse liver biopsy for histological
examination, each viscera two slices. The results
show that the results of control group and low
dose group are similar 92 days after pathological
examination. 2 mice of middle dose group are
swelling slightly on liver cells, but no
significant difference with the control group.
The livers of high dose group appear obvious
lesions and performance, like liver cells
    swelling, granular degeneration and vacuolar
degeneration. 15 days after stopping the
    medicine, pathological examination shows that
livers return to normal. Therefore, at high
doses, long-term use of Datura extracts, may
    cause damage to liver function.

6
  • 2.4. Animal renal work
  • Low dose drug group animals serum urea nitrogen
doesnt increase compared with the control group.
Middle dose group increase slightly compared to
the control group, but no statistical difference
(P gt 0.05). Animal serum urea nitrogen of high
dose group has increased significantly (P lt 0.05)
compared to the control group. The results show
that long-term medication of high doses can cause
    damage to the kidney.

7
  • 2.5 Animal blood
  • Three indicators of red cells, white cells and
platelets of mouse and rabbit have no significant
    difference by statistical treatment, indicating
that Datura stramonium has no effect on the blood
    system.
