Title: Isolation and Purification of Coenzyme Q10 and its dietary effects on reproductive physiology on mic

1
Isolation and Purification of Coenzyme Q10 and
its dietary effects on (reproductive) physiology
on mice.
  • Presented By
  • Afroza Khatun
  •                          Animal Reproductive
    Physiology Lab.
  •         23 March 2007

2
What is Coenzyme Q10?
  • Coenzyme Q10 (CoQ 10) or ubiquinone or ubiquinol
is essentially a vitamin or fat soluble
vitamin-like substance present in every cell of
the body and serves as a coenzyme for several of
the key enzymatic steps in the production of
energy within the cell. It is a biologically
active quinone and related in structure to
    vitamin K and E.
  • Chemically, CoQ10 is known as
  • 2,3-dimethyoxy-5-methyl-6-decaprenyl
  • -1,4- benzoquinone,

3
Sources of Coenzyme Q10
  • foods
  • heart, liver and kidney
  • beef, soy oil, sardines, mackerel, and peanuts.

4
Functions of the body
  • provide energy for the bodys cell growth and
    maintenance.
  • potent intracellular antioxidant which is
    important in its clinical effects.
  • prevents abnormal protein synthesis and thus age
    related degenerative disease
  • ability to transfer electrons and therefore act
    as an antioxidant
  • It has no toxicity and side effects.

5
Beneficial effects
  • heart conditions (especially heart failure)
  • muscular dystrophies
  • Parkinson's disease
  • cancer (breast cancer)
  • diabetes
  • HIV/AIDS
  • benefit the body and the Brain.
  • Mitochondrial disorders

6
Introduction
  • Coenzyme Q (CQ)- ubiquinone and lipid-soluble
    compound
  • CQ is an essential cofactor
  • CQ10 -with mitochondrial disorders
  • CQ10 improved heart muscle
  • High concentrations (CQ and VE) are found inner
    membranes of mitochondria.

7
Objectives
  • i) Isolation and purification of Coenzyme Q10 in
    different organ of mice.
  • ii) To know the dietary effect of Coenzyme Q10 on
    the different organs and  blood.
  • iii) To investigate the effect of Coenzyme Q10 on
    the reproduction on mice.
  • iv) Finally, to identify the factors which are
responsible to increase the Q10 in the body.

8
Materials and Method
  • Preparation of Diet and animals
  • 2 diets
  • Diet 1 (Control, 100 IU VE)
  • Diet 2 (500 mg CQ10/kg 100 IU VE)
  • Animals Male ICR mice (n 32 Male and Female
8 weeks old), n8 per diet

9
Sample preparation
  • 1) Blood sample
  • i) anesthetized with diethyl ether
  • ii) collected by cardiac puncture
  • iii) centrifuged at 6000?g at room temperature
    for 15 min
  • iv) Analyzed for CoQ10 concentration by HPLC
  •

10
Sample preparation
  • 2) Tissue sample
  • i) collect the liver, testis and ovary
  • ii) homogenized with distilled water
  • iii) extracted by chemicals
  • Iv)analysis for CoQ10 by HPLC.

11
  • Analysis of data
  • Data obtained will be analyzed using ANOVA
followed by Tukeys multiple comparison test.
